Influence of Synthetic Polyadenylic Polyuridylic Acid on the Production of Interferon-f and Interleukin-4 in Mice



Abstract
Synthetic polyadenylic.polyuridylic acid [poly(A). Poly(U)] is a non-toxic immunopotentiator and its antitumor activities have been recently evidenced in human cancers. However, the mechanism involved in such effect in not fully elucidated.
The purpose of this study is to determine whether poly(A).poly(U) could influence the productions of interferon(IFN)-¥ãand/or interleukin(IL)-4, both considered to play an important role in the regulation of anti-tumor effector function.
Normal or 3LL syngenic tumor-bearing(T-B) C57BL/6 mice were injected I.v. with 30¥ìg of poly(A).poly(U). Spleen cells, prepared from these mice 24h later, were cultured in the presence of 2¥ìg/ml of concanavalin A for 48h and the amounts of
IFN-¥ãand
IL-4 produced in the culture supernatants were titrated using respective ELISA kits. In parallel, NK cytotoxicities against Yac-1 target cells were measured by 4h- 51Cr release assay with the same spleen cell preparations.
@ES We have obtained the following results:
1) In vitro production of IFN-¥ãby spleen cells was significantly increased in mice treated with poly(A).poly(U) as compared to those treated with placebo. Such increments were more pronounced in T-B mice than normal controls.
2) Similarly. NK cytoxicities were also significantly enhanced in poly(A).poly(U)-treated T-B mice.
3) By contrast, the production of IL-4 was not influenced at all by poly(A).poly(U) in both groups in which IL-4 levels before the treatment were of nearly threshold for the detection.
It can be concluded that poly(A).poly(U)-induced enhancement of anti-tumor effector function might be partly due to the increased production IFN-¥ã, but not of IL-4.
